Well, I've been doing some research lately, and Grout's A History Of Western Music is an excellent resource.
From what I've found, he wrote 450 sacred motets, 102 mes, and 56 sacred madrigals.
Basically, the stepwise melody, the simplified counterpoint, being actually able to *understand* what was being sung, were things that Palestrina embodied.
in the agnus dei of the missa papae marcelli, he demonstrates a smooth melody, with each of the 5 voices harmonising in C with modulations to D,G,and A. Also different from earlier composers: over half of the root movements are tonic-dominant/dominant-tonic; he tries to systematically avoid chromatics.
another point of interest, you've probably already heard: legend has it he wrote this m to convince the Council that polyphony could sound reverent, clear, and beautiful, so that they would keep it as part of their music instead of abolishing the style; thus making Palestrina the savior of Church music.
One of the books I've been using says that's unlikely, however; the peice fits the style he used around 152 or 1563, and by then the council was pretty much over. I hope that helps.
